Obviously not. You need your pc to be turned on all the time so it can run the actual programme and host a server. Its just like a fridge turn it off and it no longer works
If you are worried about power consumption, you can always get a Raspberry PI and use that as the Pocketmine server.
How do you host a Pocketmine-MP server from a raspberry pi? I'm currently trying to find out. My model is the Raspberry Pi 2 running the Kano OS.
First you might want to boot the Raspberry from a usb drive instead an sd card, as they are more suited to long read/write performance. Sd cards might get corrupted if used for too long, like on a 24/7 server. https://www.raspberrypi.org/forums/viewtopic.php?f=29&t=44177
You need to run Raspbian. As for the SD vs. USB, why bother? SD cards are cheap and easy to backup... Just make sure you have backups and run the SD card until it dies, at that point, toss-it, and restore from backup a new one. (Or if you don't want unplanned down time replace the SD card every 6 months).

If you want to run 24/7 on a Rasberry Pie 2 I recommend going to get pocketmine for Linux and click on it, scroll all the way down for the download command. But with that money you can always buy a VPS or a hoster.
